RESUMO

Ginger (Zingiber officinale Roscoe), a member of the Zingiberaceae family, is one of the most popular spices worldwide, known since ancient times, and used both as a spice and a medicinal plant. The phenolic compounds found in ginger are predominantly gingerols, shogaols, and paradols. Gingerols are the major phenolic compounds found in fresh ginger and contain mainly 6-gingerol as well as 4-, 5-, 8-, 10-, and 12-gingerols. Gingerols possess a wide array of bioactivities, such as antioxidant and anticancer, among others. Regarding the different array of biological activities and published data on the mechanisms underlying its action, the complex interaction between three key events, including inflammation, oxidative stress, and immunity, appears to contribute to a plethora of pharmacological activities of this compound. Among these, the immunomodulatory properties of these compounds, which attract attention due to their effects on the immune system, have been the focus of many studies. Gingerols can alleviate inflammation given their ability to inhibit the activation of protein kinase B (Akt) and nuclear factor kappa B (NF-κB) signaling pathways, causing a decrease in proinflammatory and an increase in anti-inflammatory cytokines. However, given their low bioavailability, it is necessary to develop new and more effective strategies for treatment with gingerols. In order to overcome this problem, recent studies have addressed new drug delivery systems containing gingerols. In this review, the immunomodulatory activities of gingerol and its underlying mechanisms of action combined with the contributions of developed nanodrug delivery systems to this activity will be examined.

Scorzonera species belong to the Asteraceae family comprising more than 25000 species. The present study aimed to examine the phytochemical profiles and biological activities of S. sandrasica Hartvig et Strid, S. coriacea A. Duran&Aksoy, and S. ahmet-duranii Makbul&Coskuncelebi which are endemic species to Turkey. Flavonoids such as hyperoside, isoquercitrin, rutin, isoorientin, orientin, 7-O-methyl isoorientin, luteolin-7-O-ß-glycoside, apigenin-7-O-ß-glucoside, vitexin, isovitexin as well as caffeoylquinic acid derivatives including chlorogenic acid, 4,5-O-dicaffeoylquinic acid, and 1,5-O-dicaffeoylquinic acid contents were analyzed to clarify phytochemical content of the extracts. Aerial parts of the investigated extracts were determined as contain flavonoids in high amounts. Chlorogenic acid and its derivatives were detected in all investigated species, in varying amounts, both in the roots and aerial parts. S. coriacea aerial parts contained the highest total phenolic and flavonoids. The strongest inhibitory activities on ABTS and DPPH radicals were also observed with S. coriacea aerial parts by 8.07±0.28 and 13.94±0.53 µg/ml of IC50 values, respectively. Total phenolic contents of the extracts were significantly correlated with DPPH (r=-0.9842, p=0.0004) and ABTS free radical scavenging (r=-0.9870, p=0.0003) and total antioxidant capacity (r=0.8173, p=0.0470), as well as total flavonoid contents (r=0.8820, p=0.0201). S. sandrasica aerial parts and S. ahmet-duranii roots exhibited the greatest red blood cell membrane protection and protein denaturation inhibition, respectively. From the phytochemical point of view, all the selected species were analyzed for the first time.

Parkinson's Disease (PD) is a multifaceted disorder with various factors suggested to play a synergistic pathophysiological role, such as oxidative stress, autophagy, pro-inflammatory events, and neurotransmitter abnormalities. While it is crucial to discover new treatments in addition to preventing PD, recent studies have focused on determining whether nutraceuticals will exert neuroprotective actions and pharmacological functions in PD. Quercetin, a flavonol-type flavonoid, is found in many fruits and vegetables and is recognised as a complementary therapy for PD. The neuroprotective effect of quercetin is directly associated with its antioxidant activity, in addition to stimulating cellular defence against oxidative stress. Other related mechanisms are activating Sirtuins (SIRT1) and inducing autophagy, in addition to induction of Nrf2-ARE and Paraoxonase 2 (PON2). Quercetin, whose neuroprotective activity has been demonstrated in many studies, unfortunately, has a disadvantage because of its poor water solubility, chemical instability, and low oral bioavailability. It has been reported that the disadvantages of quercetin have been eliminated with nanocarriers loaded with quercetin. The role of nanotechnology and nanodelivery systems in reducing oxidative stress during PD provides an indisputable advantage. Accordingly, the present review aims to shed light on quercetin's beneficial effects and underlying mechanisms in neuroprotection. In addition, the contribution of nanodelivery systems to the neuroprotective effect of quercetin is also discussed.

Abstract The aim of this study was to determine antimicrobial activities of Alchemilla mollis, Alchemilla persica as well as ellagic acid and miquelianin against Staphylococcus aureus, Enterococcus faecalis, Bacillus subtilis, Escherichia coli, Pseudomonas aeruginosa, Candida albicans by using microbroth dilution method and anti-inflammatory activity by using human red blood cell (HRBC) membrane stabilization method. Microbroth dilution method was used to determine the antimicrobial activities. Extracts possessed activity having MIC values of 2.5-5-10mg/ mL, compounds possessed activity having MIC values of 1.25-2.5-4-5mg/mL. A.mollis aerial parts displayed the highest anti-inflammatory activity (IC50=1.22±0.07mg/mL). Ellagic acid and miquelianin were also determined as anti-inflammatory agents with 0.57±0.01mg/mL and 1.23±0.02mg/mL IC50 values, respectively. Total phenolic content and tannin content of the A.mollis and A.persica were determined as 357.00±75.80mg, 282.50±28.70mg PGE/g plant material and 18.02%, 18.63% respectively according to the method described by European Pharmacopoeia. Ellagic acid, miquelianin and catechin were analyzed by HPLC. The highest catechin content was detected in A.persica roots (6.69±0.05g/100g plant material). A.mollis aerial parts contain higher miquelianin (0.39±0.02g/plant material) and ellagic acid (1.56±0.01g/ plant material) than A.persica.

In the last five decades study on plant secondary metabolites have been increasing. Higher plants with a wide range of secondary metabolites have been very important in the search of new therapeutic agents. In this study secondary metabolites of Lathyrus armenus (Boiss. & Huet) which are endemic in Turkey, were studied. Flavonol glycosides (Rhamnocitrin-3-O-rhamninoside, Rhamnetin-3-O-rhamninoside, Rhamnazin- 3-O-rhamninoside, kaempferol3-O-rhamninoside and, kaempferol-3-O-glucosyl (1→2) rhamnoside) were isolated by different chromatographic methods and identified by 1H, 13C NMR, as well as 2D NMR and Mass spectroscopy techniques from ethyl acetate and aqueous fractions of L. armenus's methanolic extract. This is the first study about secondary metabolites of Turkish Lathyrus species.

ETHNOPHARMACOLOGICAL RELEVANCE: Scorzonera latifolia (Fisch. & Mey.) DC. (Asteraceae) grows naturally in Eastern Anatolia, northeastern Iran, and Caucasus. Latex of S. latifolia roots is used in Turkish folk medicine for its analgesic effects, externally to cure infertility in women, and internally as an antihelmintic. The milk obtained from the stem of the Scorzonera species is used for wound healing activity. Antinociceptive, anti-inflammatory, wound-healing, antioxidant, and antimicrobial activities have previously been reported for S. latifolia. AIM OF THE STUDY: A methanol extract of the aerial parts of Scorzonera latifolia that had been shown to possess wound-healing activity, was used to elucidate the possible mechanism of the wound-healing activity and to identify the compound(s) responsible for the effect by means of bioassay-guided fractionation. MATERIALS AND METHODS: The wound-healing activity potential of methanol extract of S. latifolia was detected by evaluating the inhibitory activity on the collagenase, hyaluronidase and elastase, which play important roles in the wound-healing process. Succesive fractionation of the methanol extract using petroleum ether, chloroform, ethyl acetate, respectively, and the residual wateryielded four respective fractions. The ethyl acetate part, which was determined as the most active fraction, was selected for further separation using chromatographic techniques. RESULTS: Ethylacetate fraction exhibited significant inhibitory activities on collagenase and elastase. Chromatographic separation of the ethylacetate extract yielded an active subfraction, from which was used to isolate quercetin-3-O-ß-apiofuranosyl-(1'''→2'')-ß-D-glucopyranoside (1), quercetin-3-O-α-rhamnopyranosyl-(1→6)-ß-D-galactopyranoside (2), isoorientin (3), and 7-methylisoorientin (4). Of the compounds tested, 7-methylisoorientin (4) exerted inhibitory activity on collagenase and elastase, while quercetin-3-O-ß-apiofuranosyl-(1'''→2'')-ß-glucopyranoside (1) inhibited collagenase only. None of the fractions, or isolated compounds showed any inhibitory effect on hyaluronidase. It must be mentioned, that in vitro tests showed that compounds 1-4 inhibit the collagenase and elastase and could help wound-healing process. However, the inhibititory effect of the methanol extract appears to be greater than that of both of the ethylacetate fraction, subfraction G and the isolated compounds, which suggest that a synergistic interaction of several compounds could be responsible for the wound-healing activity of the aerial parts of S. latifolia.

OBJECTIVES: This study describes the qualitative and quantitative analysis of galantamine in Sternbergia species growing in Turkey. MATERIALS AND METHODS: Galantamine was isolated from Sternbergia fischeriana bulbs and the structure of the compound elucidated by spectroscopic methods. The qualitative and quantitative analysis of galantamine was investigated in Sternbergia lutea subsp. lutea, S. lutea subsp. sicula, Sternbergia candida, S. fischeriana, and Sternbergia clusiana using a specially developed and validated high performance liquid chromatography (HPLC) method. RESULTS: S. lutea subsp. sicula had the highest content of galantamine, i.e., 0.0165±0.0002 g/100 g. The limits of detection and quantification were 7.5 µg and 25 µg, respectively. CONCLUSION: Isolation of galantamine from S. fischeriana growing in Turkey is reported for the first time. An HPLC method was developed for identification and quantification of galantamine in Sternbergia species.

Natural products are rapidly becoming the primary sources of novel antimicrobial agents, as resistance to existing antimicrobial agents is increasing. Apart from determining the antimicrobial activity of natural products, it is also important to understand their effects on the virulence factors of microorganisms. This study aimed to determine the antimicrobial activity of Sternbergia species prevalent in Turkey and investigate their role in the inhibition of germination tube and biofilm formation, both of which are known to be important virulence factors of Candida albicans. The antimicrobial activities of the plant extracts were evaluated using bore-plate and broth microdilution method. The extracts' capacity to inhibit the formation of the germ-tube was also evaluated. The findings of our study revealed that Sternbergia lutea, Sternbergia vernalis possessed antimicrobial activities, with MIC values ranging between 0.048 mg/mL and 0.39 mg/mL. The highest antimicrobial activity was observed against Candida dubliniensis (0.048 mg/mL). While evaluating the inhibition of fungal germination activities, S. vernalis extract (at a concentration of 0.09 mg/mL) was found to be the most effective against C. albicans ATCC 90028 strain. The results also indicated that S. vernalis extracts at sub-MIC levels inhibited germ tube formation and modulated the tail-length of germinated cells, both of which are important virulence factors of C. albicans. Furthermore, the inhibition of biofilm-formation was also investigated, and it was found that two Sternbergia spp. extracts at or below MIC levels inhibited biofilm formation.

In the current study, four Onobrychis species, O. albiflora Hub.-Mor., O. argyrea Boiss. subsp. argyrea Boiss., O. galegifolia Boiss., and O. tournefortii (Willd.) Desv. were collected from Anatolia to be evaluated for their antidiabetic activities. Methanol water extracts of the aerial parts were used for experiments. An alloxan-induced diabetic mice test model was used. Phytochemical analysis of the tested extracts was investigated using the HPLC method. The highest activity was observed with treatment of O. albiflora aerial part extract. Significant decrements were detected in the blood glucose levels as follows: 180.83±47.48 and 252.83±50.47mg/dL at 100 mg/kg and 200 mg/kg doses of O. albiflora, respectively, when compared to the isotonic saline solution control group, eliciting a blood glucose level of 494.20±27.32. Among the tested standard compounds, rutin and isoquercetin were detected in the examined species. The highest amount of rutin (1.1981±0.0017%) and isoquercetin (0.7318±0.0197%) were found in O. albiflora and O. argyrea subsp. argyrea, respectively. Antidiabetic activities of the tested Onobrychis species seem to indicate a possible correlation with their rutin and isoquercetin contents. Therefore, rutin and isoquercetin may be the antidiabetic compounds that contribute to the antidiabetic activity of the tested Onobrychis species.

ETHNOPHARMACOLOGICAL RELEVANCE: The latex and the aerial parts of Euphorbia characias L. (Euphorbiaceae) have been used as medicinal plant to treat wounds and warts in traditional medicine. AIM OF THE STUDY: The effect of the plant extract was tested in vivo and in vitro with experimental models to find scientific evidence for traditional use in wound healing. Potentially active wound-healer compounds were isolated from the active fraction using fractionation procedures under the guidance of biological assay and the possible role of the compounds in the wound healing process was also determined. MATERIAL AND METHODS: N-hexane, ethyl acetate, and methanol extracts were successively prepared from the aerial parts of E. characias subsp. wulfenii. The extracts were tested with linear incision, circular excision wound models and the hydroxyproline assay method to assess the wound-healing activity. The inhibition of the increase in capillary permeability induced by acetic acid, an acute inflammation model, was used to assay the anti-inflammatory activity. Different chromatographic separation techniques on sephadex and silica gel columns, and bioassay guided assay techniques have been used to isolate the active compounds of the plant. Moreover, hyaluronidase, collagenase and elastase enzymes inhibitory effect of active principle were investigated in vitro to find out the mechanism of action. RESULTS: The methanol (MeOH-ex) extract of the aerial parts of E. characias subsp. wulfenii showed significant wound healing activity (linear incision wound model: 43.04%; circular excision wound model 65.24%) and anti-inflammatory activity (34.74%). The methanol extract was separated into its fractions by column chromatography for isolation of efficient compounds. Biological activity of the fractions were assessed and further isolation and purification processes have been carried out in the active fraction. Isolation studies were carried out from the MeOH-ex fraction to obtain active constituents and their structures were elucidated to be quercetin-3-O-rhamnoside (quercitrin), quercetin-3-O-galactoside (hyperoside), and quercetin-3-O-arabinoside (guaijaverin). Further in vitro and in vivo assays showed that quercetin derivatives were responsible for the wound-healing activity of the plant, and also found to be significant anti-elastase and anti-collagenase activities. The amounts of three compounds, isolated from active fraction, were determined by using high performance liquid chromatography. Calibration equation was calculated with dilutions, prepared from pure substances, and assay was performed in total extract, prepared from E. characias subsp. wulfenii. It was detected that the plant had 1.22% quercitrin, 0.35% hyperoside, and 0.11% guaijaverin. The validation of the analytical method was performed by linearity, precision, limit of detection, and limit of quantification parameters. CONCLUSION: Present study supported the traditional use of the aerial parts E. characias subsp. wulfenii as wound healer and quercetin derivatives were isolated as active components from the active fraction by using bioassay-guided fractionation technique.

OBJECTIVES: Tanacetum species are traditionally used as insecticide, and externally wound healer as well as for anti-inflammatory and antihistaminic properties. The in vivo wound-healing and anti-inflammatory potential of four Tanacetum species, Tanacetum argenteum (Lam.) Willd. subsp. argenteum (TA), Tanacetum heterotomum (Bornm.) Grierson (TH), Tanacetum densum (Lab.) Schultz Bip. subsp. sivasicum (TD), and Tanacetum vulgare L. (TV) was investigated. MATERIALS AND METHODS: The chloroform (CHCl3) and methanol:water (80:20) extracts were prepared from the aerial parts of each plant. For assessment of the wound-healing activity, linear incision on rats and circular excision on mice wound models were used and histopathological analyses were conducted on the tissues treated with the test materials. For the evaluation of the anti-inflammatory activity, Whittle Method based on the inhibition of the acetic acid-induced increase in capillary permeability was used. In order to elucidate the phytochemical contents of the extracts, HPLC profiles of active fractions were screened and quantitative analysis was conducted within the scope of HPLC analysis. RESULTS: The CHCl3 extracts of TD, TA and TV were found to have significant wound healing activity (37.1%, 30.8% and 26.1% tensile strength; 88.05%, 72.93% and 44.88% contraction values, respectively) and anti-inflammatory activities (31.5% and 26.6% inhibition values for TD and TA). Parthenolide content of the CHCl3 extracts of TA, TH and TV were found 242.66±1.53, 190.16±5.62 and 177.51±3.73 µg/100 mg plant material, respectively. CONCLUSION: According to the results, the other secondary metabolites present in the aerial parts of the Tanacetum species possibly exerted synergistic effects on the observed healing of the wounds.

Alchemilla mollis (Buser) Rothm aerial part and root methanolic-water extracts were evaluated for their hepatoprotective activity on carbon tetrachloride induced hepatotoxicity and hypoglycemic activity on alloxan-induced diabetic mice. None of the tested extracts exhibited effects on blood glucose levels. However hepatoprotective activity results have revealed that serum ALT levels were significantly lowered by both the aerial part and root extracts at doses of 100mg/kg and 200mg/kg. Histopathological examination showed that A. mollis aerial parts and roots induced significant recovery from cellular damage; when compared to the carbon tetrachloride group, the most significant activity was observed with A. mollis aerial part extracts at a dose of 200mg/kg. There is evidence of a hepatoprotective activity of A. mollis on the phenolic content of the plant, especially in the case of flavonoids, which have potent antioxidant properties.

OBJECTIVES: The chemical composition of plants is considered to be affected by many parameters. Therefore, the region where the samples are collected is likely to have an influence on the composition of phenolic compounds, so that their biological activities. In the present study, evaluation of antioxidant activity potentials of Scorzonera latifolia (Fisch. & Mey.) DC. aerial parts and roots, which were collected from different regions of Turkey, was aimed. MATERIALS AND METHODS: 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging method and measurement of malondialdehyde (MDA) levels were used for determining antioxidant capacities of the tested extracts. In order to observe variations in the chemical composition of the investigated samples qualitatively as well as quantitatively, high performance liquid chromatography analyses were performed. RESULTS: Quantitative analysis showed that the amounts of chlorogenic acid and hyperoside in plants vary according to the regions where the samples were collected. As a result aerial parts of the S. latifolia collected from the Kars region have found to contain higher amount of chlorogenic acid (1246.78±3.20 µg/g) as well as hyperoside (652.32±2.48 µg/g) than other samples. The highest DPPH radical scavenging activity was determined with the IC50 value of 1.036 mg/mL for S. latifolia aerial parts of Kayseri sample. MDA level was detected as the lowest with treatment of S. latifolia Bayburt root sample (4.41 nmol/mL). CONCLUSION: According to the antioxidant activity results, no significant difference was observed in the antioxidant potential between the samples collected from different locations except for S. latifolia collected from the Kars region.

CONTEXT: Scorzonera L. species (Asteraceae) are edible and as medicinal plants are used for various purposed in folk medicine. OBJECTIVE: The methanol extracts of the aerial parts and roots from 27 Scorzonera taxa were investigated for their possible neurobiological effects. MATERIALS AND METHODS: Inhibitory potential of the Scorzonera species was tested against acetylcholinesterase (AChE), butyrylcholinesterase (BChE), and tyrosinase (TYRO) at 100 µg mL(-1) using ELISA microtiter assay. Antioxidant activity of the extracts was tested with radical scavenging activity, metal-chelation capacity, ferric- (FRAP), and phosphomolibdenum-reducing antioxidant power (PRAP) assays. Chlorogenic acid, hyperoside, rutin, and scorzotomentosin-4-O-ß-glucoside were also screened in the same manner. Total phenol and flavonoid quantification in the extracts were determined spectrophotometrically. RESULTS: The aerial parts of Scorzonera pisidica (40.25 ± 0.74%) and chlorogenic acid (46.97 ± 0.82%) displayed the highest TYRO inhibition, while the remaining samples showed only trivial inhibition against cholinesterases (2.08 ± 1.35%-25.32 ± 1.37%). The same extract of S. pisidica was revealed to be the most potent in scavenging of all three radicals and FRAP assay. DISCUSSION AND CONCLUSION: Out of 27 taxa, S. pisidica, in particular, may deserve further investigation for its neuroprotective potential.

Wound healing and anti-inflammatory effects of aerial parts and roots of S. acuminata, S. cana var. alpina, S. cana var. jacquiniana, S. cana var. radicosa, S. eriophora, S. laciniata ssp. laciniata, S. suberosa ssp. suberosa and S. sublanata were investigated in current study to clarify the traditional usage of Scorzonera species growing in Turkey. It is well known that some species of the Scorzonera genus are used for wound healing in Turkish and European traditional medicine. Therefore, wound healing effect of the plants was assessed by using linear incision and circular excision experimental wound models and subsequently histopathological analysis. Hydroxyproline content of the treated tissues was also assessed. Furthermore, the extracts were screened for anti-hyaluronidase activity. For the evaluation of anti-inflammatory activity, acetic acid-induced increase in capillary permeability test was used. 20% aqueous methanol extracts of the aerial parts of Scorzonera species, S. cana var. jacquiniana and S. eriophora were found to be effective on the wound and anti-inflammatory activity test models. The results of histopathological examination supported the outcome of linear incision and circular excision wound models. Phytochemical analyses of the tested extracts were also performed by using developed and validated HPLC method. Among the tested standard compounds, varying amounts of the chlorogenic acid, rutin, hyperoside and luteolin-7-glycoside were determined in Scorzonera species.

RESUMO

A novel triterpene 1 (3-ß-hydroxy-fern-7-en-6-one-acetate) together with four known compounds, urs-12-en-11-one-3-acetyl (2), 3-ß-hydroxy-fern-8-en-7-one-acetate (3), olean-12-en-11-one-3-acetyl (4) and leucodin (5) were obtained from the S. latifolia roots. All compounds were isolated from the n-hexane extract of S. latifolia roots using several chromatographic techniques. The structure of the isolated compounds was elucidated on the basis of (1)H-NMR, (13)C-NMR and 2D NMR data (HMBC, HMQC, COSY, TOCSY, NOESY, DEPT) as well as GC EITOF-HRMS.

ETHNOPHARMACOLOGICAL RELEVANCE: Scorzonera species are mainly used against arteriosclerosis, kidney diseases, hypertension, diabetes mellitus and rheumatism, as well as for pain relief and wound healing in Turkish folk medicine. In the current study, aerial parts and roots of Scorzonera cinerea, Scorzonera incisa, Scorzonera latifolia, Scorzonera mollis ssp. szowitsii, Scorzonera parviflora, and Scorzonera tomentosa collected from different regions of Anatolia and yaki sakizi which is prepared by drying the latex is obtained from the roots of Scorzonera latifolia were evaluated for their wound healing and anti-inflammatory effects. MATERIALS AND METHODS: In vivo wound healing activity of the plants was evaluated by linear incision and circular excision wound models. Hydroxyproline content of the treated tissues was also assessed. Acetic acid-induced capillary permeability test was used for anti-inflammatory activity. For the determination of the active constituents, a new high performance liquid chromatographic method was developed. Precision of the developed method was performed by the evaluation of intra-day and inter-day variations of the each standard at limits of quantification (LOQ) levels. RESULTS: The aqueous methanolic extracts of Scorzonera latifolia, Scorzonera mollis ssp. szowitsii demonstrated the highest activities. The main compound was detected as chlorogenic acid both in roots and aerial parts of the most Scorzonera species and hyperoside or rutin was determined as one of the main component of the aerial parts. CONCLUSION: The experimental study revealed that methanol-water extracts from aerial parts of Scorzonera latifolia and Scorzonera mollis ssp. szowitsii were found to have better activity on the wound healing compared to the other extracts and control groups. This might be due to the combined effect of the constituents present in the extracts.
